Abstract

Penelitian ini bertujuan untuk menganalisis intensitas pewarnaan, luas area positif, dan jumlah sel positif Tumor Necrosis Factor-Alpha (TNF-α) pada jaringan pulmo tikus Wistar diabetik yang diberi ekstrak daun Tithonia diversifolia. Penelitian menggunakan metode true experimental dengan rancangan Post Test Only Control Design pada 18 ekor tikus Wistar yang dibagi menjadi tiga kelompok, yaitu kontrol normal (IKL), kontrol diabetes tanpa perlakuan (IT4), dan kelompok diabetes yang diberi ekstrak Tithonia diversifolia (IT8). Analisis ekspresi TNF-α dilakukan menggunakan metode imunohistokimia dengan sistem skoring semi-kuantitatif berdasarkan intensitas pewarnaan dan jumlah sel imunopositif pada lima lapang pandang pembesaran 400×. Hasil penelitian menunjukkan bahwa kelompok IT4 memiliki rata-rata skor IRS tertinggi sebesar 6,33, diikuti kelompok IT8 sebesar 3,87, dan kelompok IKL sebesar 1,60. Analisis statistik menggunakan uji Kruskal-Wallis menunjukkan adanya perbedaan yang signifikan antar kelompok (p < 0,05). Pemberian ekstrak daun Tithonia diversifolia terbukti mampu menurunkan ekspresi TNF-α pada jaringan pulmo tikus Wistar diabetik.
