Abstract

Penelitian ini bertujuan untuk menganalisis imunoekspresi Cyclooxygenase-2 (COX-2) pada jaringan hati tikus Sprague Dawley setelah pemberian fermentasi buah berenuk (Crescentia cujete L.). Penelitian ini merupakan penelitian eksperimental menggunakan 16 ekor tikus jantan galur Sprague Dawley yang dibagi ke dalam dua kelompok, masing-masing terdiri atas 8 ekor, yaitu kelompok kontrol (P0) dan kelompok perlakuan (P1) yang diberikan fermentasi buah berenuk dosis 500 mg/kg BB secara oral. Seluruh hewan coba terlebih dahulu menjalani masa adaptasi selama 7 hari sebelum perlakuan dilakukan. Pada hari ke-91, jaringan hati diambil untuk pemeriksaan imunohistokimia guna mengetahui tingkat imunoreaktivitas COX-2, kemudian data dianalisis menggunakan uji t independen. Hasil penelitian menunjukkan bahwa pemberian fermentasi buah berenuk tidak memberikan pengaruh yang signifikan terhadap imunoekspresi COX-2 pada hati tikus dengan nilai p = 0,59 (p > 0,05). Rerata ekspresi COX-2 pada kelompok kontrol sebesar 72,88%, sedangkan pada kelompok perlakuan sebesar 74,48%. Meskipun terdapat kecenderungan peningkatan ekspresi COX-2 pada kelompok perlakuan, perbedaan tersebut tidak bermakna secara statistik. Temuan ini menunjukkan bahwa pemberian fermentasi buah berenuk dosis 500 mg/kg BB belum menyebabkan peningkatan respons inflamasi yang signifikan pada jaringan hati, sehingga diduga hati masih mampu mempertahankan mekanisme adaptasi fisiologis terhadap paparan fermentasi buah berenuk.
