Abstract

Penelitian ini bertujuan untuk mengidentifikasi keberadaan Salmonella sp. dan menganalisis gambaran histopatologi pada limpa ayam broiler (Gallus domesticus) yang terinfeksi salmonellosis. Sampel diambil dari pasar tradisional Surabaya menggunakan metode purposive sampling, terdiri dari ayam yang bergejala dan tidak bergejala salmonellosis. Isolasi bakteri dilakukan melalui pre-enrichment dan enrichment menggunakan media Buffered Peptone Water (BPW) dan Rappaport Vassiliadis (RV), dilanjutkan dengan isolasi pada media Xylose Lysine Deoxycholate (XLD). Identifikasi bakteri dilakukan melalui pewarnaan Gram dan uji biokimia TSIA, SIM, urease, MR, dan VP. Pemeriksaan histopatologi dilakukan pada preparat limpa yang diberi pewarnaan Hematoxylin Eosin (HE) dan dianalisis secara deskriptif dengan skoring nekrosis, hemoragi, dan degenerasi. Hasil penelitian menunjukkan bahwa ayam broiler yang bergejala salmonellosis positif mengandung Salmonella sp., sedangkan ayam yang tidak bergejala negatif. Limpa ayam yang terinfeksi menunjukkan gambaran histopatologi berupa pembesaran folikel limfoid, kongesti pembuluh darah, infiltrasi sel radang, nekrosis fokal, hemoragi, dan degenerasi sel. Penelitian ini dapat menjadi dasar diagnosis mikroskopis salmonellosis pada ayam broiler dan meningkatkan pengendalian infeksi melalui manajemen pemeliharaan yang lebih baik.
