Abstract
Goat meat is a food source of animal protein that is easily damaged by the growth of microorganisms. This study aims to determine the effect of administering turmeric leaf simplicia (Curcuma domestica) on the total number of bacteria and Escherichia coli contamination in goat meat. The study used a laboratory experimental method with a Completely Randomized Design (CRD) consisting of five treatments and five replications, namely P0 (-) without treatment, P0 (+) stored for 1 hour at room temperature, P1 given turmeric leaf simplicia and stored for 1 hour, P2 given turmeric leaf simplicia and stored for 1.5 hours, and P3 given turmeric leaf simplicia and stored for 2 hours. The parameters observed included the total number of bacteria using the Total Plate Count (TPC) method and identification of Escherichia coli through EMBA media, Gram staining, and biochemical tests. The results showed that the administration of turmeric leaf simplicia was able to reduce the total number of bacteria in goat meat, with the average TPC values sequentially at P0(-), P0(+), P1, P2, and P3 of 81.25 × 10⁴ CFU/g, 105.12 × 10⁴ CFU/g, 29.3 × 10⁴ CFU/g, 2.06 × 10⁴ CFU/g, and 16.38 × 10⁴ CFU/g. The results of the ANOVA analysis showed a significant difference between treatments (p<0.05). However, all samples still showed positive results for Escherichia coli contamination based on the growth of metallic green colonies on EMBA media, the results of Gram-negative rod-shaped staining, and biochemical tests. Thus, it can be concluded that turmeric leaf (Curcuma domestica) simplicia is effective in reducing the total number of bacteria in goat meat, especially in the 1.5 hour storage treatment, but has not been able to eliminate Escherichia coli contamination completely.
