Abstract
Fermentasi buah berenuk (Crescentia cujete L.) diketahui mengandung berbagai senyawa bioaktif, termasuk kolin, flavonoid, saponin, dan tanin yang berpotensi memberikan efek protektif terhadap jaringan. Meskipun demikian, keamanan penggunaan jangka panjang fermentasi buah berenuk masih perlu dievaluasi melalui uji toksisitas subkronik. Penelitian ini bertujuan untuk mengetahui pengaruh paparan subkronik selama 90 hari fermentasi buah berenuk dosis 500 mg/kg berat badan terhadap imunoekspresi Vascular Endothelial Growth Factor (VEGF) pada lambung tikus Sprague Dawley. Penelitian ini menggunakan 16 ekor tikus putih jantan Sprague Dawley yang dibagi menjadi dua kelompok, yaitu kelompok kontrol (P0) dan kelompok perlakuan (P1). Kelompok perlakuan diberikan fermentasi buah berenuk dosis 500 mg/kg BB secara oral selama 90 hari, sedangkan kelompok kontrol tidak diberikan perlakuan. Pada hari ke-91 dilakukan nekropsi dan pengambilan organ lambung untuk pemeriksaan imunohistokimia VEGF. Data hasil persentase area positif VEGF dianalisis menggunakan uji Independent Sample t-test dengan tingkat kepercayaan 95%. Hasil penelitian menunjukkan nilai rerata ekspresi VEGF pada kelompok kontrol sebesar 13,95%, sedangkan pada kelompok perlakuan sebesar 11,12%. Analisis statistik menunjukkan tidak terdapat perbedaan yang signifikan antara kedua kelompok (p = 0,121; p > 0,05). Berdasarkan metode skoring imunohistokimia, kedua kelompok berada pada kategori skor 1 (intensitas lemah). Hasil tersebut menunjukkan bahwa pemberian fermentasi buah berenuk dosis 500 mg/kg BB selama 90 hari tidak meningkatkan ekspresi VEGF pada jaringan lambung tikus Sprague Dawley. Dengan demikian, fermentasi buah berenuk pada dosis tersebut tidak menunjukkan efek toksik terhadap lambung dan berpotensi aman untuk dikembangkan sebagai produk herbal.

However, the long-term safety of fermented calabash fruit remains to be evaluated through subchronic toxicity studies. This study aimed to determine the effect of a 90-day subchronic exposure to fermented calabash fruit at a dose of 500 mg/kg body weight on the immunoexpression of Vascular Endothelial Growth Factor (VEGF) in the stomach of Sprague Dawley rats. A total of 16 male Sprague Dawley rats were randomly divided into two groups: a control group (P0) and a treatment group (P1). The treatment group received fermented calabash fruit orally at a dose of 500 mg/kg body weight for 90 consecutive days, while the control group received no treatment. On day 91, the rats were euthanized, and stomach tissues were collected for VEGF immunohistochemical examination. The percentage of VEGF-positive areas was analyzed using an Independent Samples t-test at a 95% confidence level. The results showed that the mean VEGF expression was 13.95% in the control group and 11.12% in the treatment group. Statistical analysis revealed no significant difference between the two groups (p = 0.121; p > 0.05). Based on the immunohistochemical scoring method, both groups were classified as score 1 (weak intensity). These findings indicate that administration of fermented calabash fruit at a dose of 500 mg/kg body weight for 90 days did not increase VEGF expression in the gastric tissue of Sprague Dawley rats.
