Pembimbing: Anna LewiSantoso, dr., M.Si Padapenderita diabetes mellitus tipe I ditemukanperubahan-perubahanpadapankreasberupapengecilanukurandaripankreas, atrofipadabagianeksokrinpankreas, danatrofisel-selasinar di sekitarpulau Langerhans yang mengalamidegenerasi, sedangkanpada diabetes mellitus Tipe II yang terjadiadalahketidakseimbangandarisekresieksokrinpankreasdangangguankontrolglukosadarah. Hiperglikemipada diabetes melitusdapatmenyebabkanaktivasijalurmetabolismepoliolsehinggameningkatkanpembentukansenyawaoksigenreaktif (ROS).ROS yang berlebihandapatmenyebabkanstresoksidatifdandapatmemperparahkerusakansel beta pankreas.Dari latarbelakangtersebutmakapenelitianinidilakukandengantujuanuntukmengetahuipengaruhpemberianekstraketanolkulitbuahmanggis(Garciniamangostana L.) terhadapgambaranhistologiluaspulau Langerhans tikusputihjantankondisihiperglikemik. Populasidalampenelitianiniadalahtikuswistarputih (Rattusnorvegicus) jantandalamkondisisehat yang berumur 2-3 bulandenganberatbadankuranglebih 150-200 gram denganjumlahsampel 28 ekor yang terbagidalam 4 kelompok. Variabelbebaspenelitianiniadalahdosisekstraketanolkulitbuahmanggisdanvariabelterikatnyaadalahluaspulau Langerhans pankreas.Tikusmenjadihiperglikemikdenganpemberianaloksan 80 mg/kgBBintraperitoneal yang diberikanpadakelompok K+, P1 dan P2.Kelompok P1 diberiekstraketanolkulitbuahmanggis 250 mg/kgBB/haridankelompok P2 500 mg/kgBB/hariselama 14 hari. Analisis data dalampenelitianinimenggunakanujiOne Way ANOVA.Dari hasilpenelitianmenunjukkanadanyapengaruhpemberianekstraketanolkulitbuahmanggisterhadapgambaranhistologiluaspulau Langerhanstikusputihjantankondisihiperglikemik.Hasilpenelitianjugamenunjukkanbahwapadatikus yang diberiekstraketanolkulitbuahmanggismengalamipeningkatanpadaluaspulaulangerhansnya.Luaspulaulangerhanstikusputihjantankondisihiperglikemik yang diberiekstraketanolkulitbuahmanggis 250 mg/kgBB/hariadalahsebesar 10,83, sedangkanluaspulaulangerhanstikusputihjantankondisihiperglikemik yang diberiekstraketanolkulitbuahmanggis 500 mg/kgBB/hariadalahsebesar 11,11. The Effect of Giving Mangosteen Fruit Leather Ethanol Extract (Garciniamangostana L.) on Overview of Histology Area of Langerhans Island on White Male Mice that are in Hyperglycemic Condition. Si In patients with type I diabetes mellitus found changes in the pancreas in the form of reducing the size of the pancreas, atrophy in the exocrine pancreas, and atrophy of acinar cells around the degenerated Langerhans island, whereas in Type II diabetes mellitus, changes that occurs are an imbalance of secretion exocrine pancreas and impaired blood glucose control. Hyperglycemia in diabetes mellitus may lead to activation of the polyol metabolism pathway thus increasing the formation of reactive oxygen (ROS) compounds. Excessive ROS can cause oxidative stress and can aggravate the destruction of pancreatic beta cells. From this background, this research was conducted with the aim to know the effect of ethanol extract of mangosteen peel (Garciniamangostana L.) to the overview of histology area of Langerhans island on white male mice that is in hyperglycemic condition. The population in this study was male white wistar mice (Rattusnorvegicus) in healthy condition aged 2-3 months with body weight of approximately 150-200 gram with sample number 28 tails divided into 4 groups. The independent variables of this study were the dosage of ethanol extract of mangosteen peel and the dependent variable was the area of Langerhans island of pancreas. The mice became hyperglycemic with intraperitoneal 80 mg / kgBBalloxan administration given to the K +, P1 and P2 groups. The P1 group was extracted with 250 mg / kg / day and 500 mg / kg / day of manganese ethanol extract for P2 group for 14 days. Data analysis in this research using One Way ANOVA test. From the results of the research showed that there is an effect of giving ethanol extract of mangosteen peel to the broad histology of Langerhans island of white male mice that is in hyperglycemic condition. The results also showed that in mice that was given ethanol extract of mangosteen peel, the wide of its island langerhans has increased. The area of Langerhans Island on white male mice which has hyperglycemic condition that given ethanol extract of mangosteen peel of 250 mg / kgBW / day is 10.83 micrometers, whereas the area of Langerhans Island on white male mice which has hyperglycemic condition that given ethanol extract of mangosteen peel of 500 mg / kgBW / day is 11.11 micrometers.
