Abstract

This study was conducted to identify plaque types and measure plaque size of bacteriophages isolated from swiftlet houses. Testing for lytic and lysogenic bacteriophage activity was carried out using a series of methods: subculture on nutrient agar (NA), overnight culture, enrichment isolation, and plaque assay. Plaque type and size were observed using a monocular digital microscope, and data were analyzed descriptively to describe the characteristics of bacteriophage lytic activity on host cells. The results showed that of the 12 bacteriophage isolates from swiftlet house fecal waste, 7 phage codes formed clear lysis zones. The plaque morphology of each isolate code was consistent, characterized by a generally round plaque shape, uniform edges, and relatively small variations in diameter, indicating uniform phage characteristics within each isolate.
