Abstract
GAMBARAN HISTOPATOLOGI GINJAL YANG DIINDUKSI CISPLATIN DAN EKSTRAK DAUN BANGUN-BANGUN (Coleus ambonicus, L.) DENGAN PEWARNAAN MALLORY AZAN PADA TIKUS PUTIH (Rattus norvegicus) Melda Putri Elizabeth Supriyanto ABSTRAK Penelitian ini bertujuan untuk menilai efek dari induksi Cisplatin dan ekstrak Daun Bangun-bangun (Coleus amboinicus, L.) terhadap gambaran hemoragi, kongesti, dan vaskulitis pada ginjal tikus putih (Rattus norvegicus) dengan pewarnaan Mallory azan. Hewan coba yang digunakan adalah tikus putih sejumlah 18 ekor. Rancangan yang digunakan adalah rancangan acak lengkap dengan 3 perlakuan dan 6 kali ulangan. Ketiga perlakuan tersebut adalah P0 (kontrol), P1 (induksi cisplatin 7mg/kgBB) pada hari keempat secara intraperitonial, suspensi CMC NA 1% setiap hari, dan P2 (diberi ekstrak Daun Bangun-bangun (Coleus ambonicus, L) 100mg/kgBB secara peroral selama setiap hari dan pada hari keempat diinduksi cisplatin 7mg/kgBB). Pembuatan histopat menggunakan pewarnaan Mallory Azan. Data yang di peroleh dianalisis menggunakan uji Kruskal Wallis kemudian dilanjutkan dengan uji Mann Whitney Test, kongesti (p<0,05), hemoragi (p<0,05), vaskulitis (p<0,05). Hasil uji Mann Whitney Test menunjukkan terdapat perbedaan antar kelompok (p<0,05) berarti pemberian ekstrak Daun Bangun-bangun dengan dosis 100g/KgBB berbeda nyata (signifikan) dengan yang hanya diinduksi cisplatin 7mg/kgBB. Hasil penelitian menunjukkan bahwa pemberian ekstrak Daun Bangunbangun (Coleus ambonicus, L) mampu menurunkan jumlah kerusakan hemoragi, kongesti dan vaskulitis. KIDNEY HISTOPATHOLOGIC FEATURES INDUCED BY CISPLATIN AND BANGUN’S LEAF EXTRACT (Coleus ambonicus, L) WITH MALLORY AZAN STAINING IN WHITE RAT (Rattus norvegicus) Melda Putri Elizabeth Supriyanto ABSTRACT This study aimed to assess the effect of Cisplatin induction and Bangun’s Leaf extract (Coleus amboinicus, L.) on the histopathological picture of kidney white rats (Rattus norvegicus) hemorhage, congestion, and vasculitis with Mallory Azan staining method. A total of 18 rats were used for this study. The design used in this research was a completely randomized design with 3 treatments and 6 repetitions. The treatments were P0 (control), P1 (induced by 7mg/kg body weight of cisplatin) on the fourth day intraperitonially, CMC NA 1% suspension every day., and P2 (given 100mg/kg body weight of Bangun’s Leaf extract (Coleus ambonicus, L.) orally every day and on the fourth day induced by 7mg/kg body weight of cisplatin). Histophatology stain using Mallory Azan staining method. The data was obtained and analyzed using the of Kruskal Wallis Test. Then followed by the Mann Whitney Test. The results of Kruskal Wallis Test, hemorrhagic (p<0,05), congestion (p<0,05), vasculitis (p<0,05) was significant. The results of Mann Whitney Test showed that there were differences between groups (p<0,05), that mean the given of Bangun’s Leaf extract (Coleus ambonicus, L.) at a dose of 100mg/kg body weight was significantly different was the other group which only given by cisplatin at dose 7mg/kg body weight. The result showed that the given of Bangun’s Leaf extract (Coleus ambonicus, L) can reduce the damage of hemorhage, congestion, and vaskulitis.
