Abstract
ABSTRAK Penelitian ini dilakukan untuk menghitung total koloni bakteri dengan menggunakan Total Plate Count (TPC) dan untuk mengidentifikasi bakteri Salmonella sp. dengan menggunakan media Salmonella-Shigella Agar (SSA), pewarnaan Gram, dan uji Triple Sugar Iron Agar (TSIA) pada ikan nila (Oreochromis niloticus). Sebanyak 30 ekor ikan nila (Oreochromis niloticus) diambil dari tambak Sawiyoh Sidoarjo untuk diperiksa di Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Wijaya Kusuma Surabaya. Penelitian ini merupakan penelitian deskriptif observasional. Hasil dari penelitian ini, dalam penghitungan Total Plate Count (TPC) menunjukkan hasil rata-rata koloni bakteri 15,8 x 105 CFU/gram, selain itu dari 30 sampel ikan nila (Oreochromis niloticus) yang telah diperiksa menunjukkan adanya suspect bakteri Salmonella sp. dengan ciri-ciri koloni yang tumbuh pada media SSA (SalmonellaShigella Agar) adalah koloni tidak berwarna dengan titik hitam di tengahnya. Pewarnaan Gram pada 30 sampel ikan nila (Oreochromis niloticus) menunjukkan hasil bakteri Gram negatif serta berbentuk batang, sedangkan pada uji TSIA (Triple Sugar Iron Agar) didapatkan satu sampel positif bakteri Salmonella sp. dengan ciri-ciri bagian slant media berwarna merah, bagian butt media berwarna kuning, dan terdapat hydrogen sulfida (H2S). Berdasarkan rangkaian penelitian yang telah dilakukan, diketahui bahwa uji TSIA (Triple Sugar Iron Agar) memiliki kedudukan tertinggi dalam penelitian ini, dikarenakan uji TSIA (Triple Sugar Iron Agar) merupakan uji gula-gula lengkap dan juga dapat menunjukkan pembentukan hydrogen sulfida (H2S), sehingga hasil dari uji TSIA (Triple Sugar Iron Agar) ini merupakan hasil akhir dari deteksi bakteri Salmonella sp. pada ikan nila (Orechromisniloticus). Kata kunci: Ikan Nila (Oreochromis niloticus), Salmonella sp., TPC (Total Plate Count), Pewarnaan Gram, TSIA (Triple Sugar Iron Agar) ABSTRACT This study was conducted to calculate the total bacterial colonies using Total Plate Count (TPC) and to identify Salmonella sp. bacteria by using Salmonella-Shigella Agar (SSA) as the media, Gram staining, and Triple Sugar Iron Agar (TSIA) test on tilapia (Oreochromis niloticus). A total of 30 tilapia (Oreochromis niloticus) were taken from the Sawiyoh Sidoarjo pond for examination at the Microbiology Laboratory of the Faculty of Medicine, Wijaya Kusuma University, Surabaya. This research was an observational descriptive study. The results of this study, in calculating the Total Plate Count (TPC) showed the results of the average bacterial colonies was 15.8 x 105 CFU/gram, besides that, from 30 samples of tilapia (Oreochromis niloticus) that had been examined showed suspect Salmonella sp. with the characteristics of colonies that grow on SSA (Salmonella-Shigella Agar) media were colorless colonies with black dots in the middle. Gram staining in 30 samples of tilapia (Oreochromis niloticus) showed the results of Gram negative bacteria and rod-shaped, whereas in the TSIA (Triple Sugar Iron Agar) test showed one positive sample of Salmonella sp. with the characteristics of the slant part of the media was red, the butt part of the media was yellow, and there was also hydrogen sulfide (H2S). Based on a series of studies that have been conducted, it is known that the TSIA (Triple Sugar Iron Agar) test has the highest position in this study, because the TSIA (Triple Sugar Iron Agar) test was a complete confection test and can also show the formation of hydrogen sulfide (H2S), so this result of the TSIA (Triple Sugar Iron Agar) test is the final result of the detection of Salmonella sp. in tilapia (Orechromis niloticus).
