Abstract
Calabash fruit (Crescentia cujete L.) contains bioactive compounds with potential biological effects; however, long-term exposure requires comprehensive safety evaluation. Repeated administration of fermented calabash fruit may influence gastrointestinal inflammatory responses, including alterations in cyclooxygenase-2 (COX-2) immunoexpression in the duodenum. The study aimed to evaluate the effect of 90-day subchronic exposure to fermented calabash fruit at a dose of 500 mg/kg body weight on COX-2 immunoexpression in the duodenum of Sprague Dawley rats. An experimental labaratory study was conducted using a 90-day subchronic toxicity design, in which Sprague Dawley rats were assigned to control and treatment groups, followed by histopathological examination and immunohistochemical analysis of COX-2 expression in duodenal tissue. The results showed that the mean percentage of COX-2 positive area was 7.63 ± 3.20% in the control group and 5.50 ± 1.27% in the treatment group, with no. statistically significant difference between groups (p>0,05). The treatment group showed a lower mean COX-2 immunoexpression compared to the control group. In conclusion, administration of fermented calabash fruit at 500 mg/kg body weight for 90 days did not increase inflammatory response in the duodenum of Sprague Dawley rats, indicating a favorable safety profile for duodenal tissue.
