Abstract
Fermentation of berenuk fruit (Crescentia cujete) is known to contain various bioactive compounds with potential anti-inflammatory and neuroprotective effects. One indicator of inflammation in brain tissue is the expression of Cyclooxygenase- 2 (COX-2), which plays a role in neuroinflammatory processes. This study aimed to determine the effect of subchronic exposure for 90 days to fermented berenuk fruit at a dose of 500 mg/kg body weight on COX-2 immunoexpression in the brains of Sprague Dawley rats. The study used 16 male Sprague Dawley rats, randomly divided into two groups: the control group (P1) and the treatment group (P2). The treatment group received fermented berenuk fruit at a dose of 500 mg/kg BW orally for 90 days, while the control group received only distilled water. On day 91, brain organ collection and immunohistochemistry examination were performed using anti-COX-2 antibodies. Data were analyzed using the Independent Sample t-test with a 95% confidence level. The results showed that the mean COX-2 immunoexpression in the control group was 7.44 ± 1.53%, while in the treatment group it was 6.32 ± 1.68%. Statistical analysis indicated no significant difference between the two groups (p > 0.05). Nevertheless, the treatment group showed a tendency toward decreased COX-2 expression compared to the control group. Based on these findings, it can be concluded that subchronic exposure to fermented berenuk fruit at a dose of 500 mg/kg BW for 90 days does not have a significant effect on COX-2 immunoexpression in the brains of Sprague Dawley rats, although it shows a tendency to reduce COX-2 expression.
