Abstract

ABSTRAK Penelitian ini dilakukan untuk mengetahui keberadaan bakteri Escherichia coli serta mengetahui total bakteri yang berada pada feses cicak rumah. Sejumlah 30 ekor cicak rumah diambil dari penjual cicak di Surabaya dengan kriteria SVL 4- 6cm dan tinggal di kawasan indoor perumahan. Sampel cicak dibawa menuju Laboratorium Mikrobiologi Fakultas Kedokteran Hewan Universitas Wijaya Kusuma Surabaya untuk diperiksa. Sampel yang digunakan ialah feses cicak segar sejumlah 0,1 gram. Pemeriksaan bakteri Escherichia coli dilakukan dengan penanaman bakteri pada media agar EMBA dengan metode streak, pemeriksaan mikroskop dengan perbesaran 1000x, dilanjutkan dengan Uji Biokimia indol, methyl red, voges proskauer, sitrat (IMViC) serta Triple Sugar Iron Agar (TSIA). Sedangkan pemeriksaan Total Plate Count feses cicak dilakukan dengan penanaman bakteri menggunakan Nutrient agar pada pengenceran 105 dan 106 dengan metode tuang kemudian dihitung secara manual dan diolah hasil datanya menggunakan rumus TPC. Hasil penelitian menunjukan bahwa 5 dari 30 cicak rumah positif bakteri Escherichia coli dan memiliki total rata-rata TPC sebesar 2x108 CFU/ml. Kata kunci: Escherichia coli, cicak rumah, feses, total plate count ABSTRACT This research was conducted to find out the existence of Escherichia coli and total bacteria in house gecko’s feces. 30 house geckos were taken from house gecko seller in Surabaya with requirements of SVL 4 to 6cm and live indoor. House gecko samples brought to Microbiology Laboratorium of Wijaya Kusuma Surabaya University to be examined. 0,1gram of fresh house gecko feces were used as samples. Escherichia coli detection was conducted with bacteria isolation in EMB agar using streak method, microscope examination with 1000x magnification, and biochemical test such as indole, methyl red, voges proskauer, citrate (IMViC), and Triple Sugar Iron Agar (TSIA). While Total Plate Count of house gecko feces were conducted with bacteria with dilution of 105 and 106 isolated in nutrient agar using pour plate method. The results were counted manually and processed with TPC formula. Research result shows that 5 out of 30 house geckos have Escherichia coli bacteria and average of Total plate count are 2x108 CFU/ml.
