Abstract

WHO in 2019 reported that at least 700,000 people worldwide died from antibiotic resistance. Staphylococcus aureus bacteria is one of the bacteria that most often becomes resistant to antibiotics. One of the alternative is using medicinal plants as natural antibacterials in preventing the increasing antibiotic resistance. Robusta coffee is a plant that has several antibacterial compounds such as caffeine, chlorogenic acid, trigonelin, polyphenols, tocopherols, and diterpenes. The aim of this study was to determine bactericidal effect of the robusta coffee bean extract in the growth of Staphylococcus aureus bacteria with dilution method. Robusta coffee bean extract divided into 5 concentrations, which were the concentrations of 20%, 40%, 60%, 80%, and 100%. After that, the extract mixed with a suspension of Staphylococcus aureus bacteria and incubated for 24 hours. The mixture was then inoculated on NAP media and observed. Data analysis used visual descriptive analysis based on the presence or absence of colonies growth on NAP media. The results showed that the higher the concentration, the stronger the bactericidal effect. Concentrations of 20% and 40% extract do not have bactericidal effect against Staphylococcus aureus. The Minimal Bactericidal Concentration (MBC) was obtained in concentration of 60% indicated by the absence of colonies growth on the NAP media. Concentrations of 80% and 100% also have bactericidal effect against Staphylococcus aureus. The conclusion of this study is that robusta coffee bean extract has the bactericidal effect in the growth of Staphylococcus aureus.
