Abstract

ABSTRAK Penelitian ini bertujuan untuk mengetahui kelayakan larva Black Soldier Fly (Hermetia illucens) yang digunakan sebagai bahan pakan ditinjau dari total kuman dan kandungan Eshcherichia coli. Larva Black Soldier Fly (BSF) yang digunakan berasal dari media limbah sayuran dan buah-buahan. Sampel Larva Black Soldier Fly (BSF) yang digunakan sebanyak 30 sampel dimana 15 sampel pada masing-masing media. Rancangan yang digunakan adalah rancangan acak lengkap. Sampel larva akan diuji dengan uji Total Plate count (TPC) dan uji kandungan Eshcherichia colidengan menggunakan media Eosin Methylen Blue Agar (EMBA). Untuk uji TPC sampel larva dilakukan pengencaran dari 10-1sampai 10-5 kemudian di inokulasikan pada media Nutrient agar (NA) dan di inkubasi selama 24 jam. Pengujian kandungan Eshcherichia colidilakukan dengan menginokulasikan suspensi larva ke media Eosin Methylen Blue Agar (EMBA) dan di inkubasi selama 24 jam. Hasil dari uji TPC menunjukkan bahwa jumlah koloni yang tumbuh melebihi batas maksimum SNI cemaran mikroba pada bahan pakan. Koloni bakteri yang tumbuh pada media Eosin Methylen Blue Agar (EMBA) tidak menunjukkan bakteri Eshcherichia coli. Hasil pemeriksaan mikroskopis menunjukkan bakteri berbentuk batang, berwarna ungu (gram positif) dan pada bagian tengah berwarna transparan yang menujukkan spora. Kata Kunci : Larva Black Soldier Fly (BSF), Eshcherichia coli, dan total kuman ABSTRACT The study aimed to determine the feasibility of Black Soldier Fly larvae (Hermetia illucens) which used as feed ingredient by total germs and Escherichia coli content observed. The Black Soldier Fly (BSF) larvae obtained from vegetables and fruits waste. Black Soldier Fly (BSF) samples used were 30 samples which 15 were sampled in each media. The design used was a full-randomized design. Larva samples were examined by Total Plate count (TPC) test and Escherichia coli content test using Eosin Methylene Blue Agar (EMBA) media. For the TPC test, the larvae samples were diluted from 10-1 to 10-5 then inoculated on Nutrient Agar (NA) media and incubated for 24 hours. Escherichia coli content was examined by inoculating the suspension of larvae into Eosin Methylene Blue Agar (EMBA) media and they were incubated for 24 hours. The results of the TPC test shows that the number of growing colonies exceed the maximum limit of SNI for microbial contamination in feed ingredients. Bacterial colonies that grow on Eosin Methylene Blue Agar (EMBA) media not Escherichia coli bacteria. The result of microscopic examination shows that the bacteria is rod-shaped, purple-colored (gram-positive) and have a transparent core which represents the spores.
