Abstract

Pajanan asap rokok mengandung berbagai senyawa toksik yang dapat memicu stres oksidatif dan respons inflamasi, sehingga berperan dalam kerusakan fungsi maupun struktur ginjal. Kondisi ini dapat ditunjukkan oleh peningkatan kadar blood urea nitrogen (BUN), kreatinin, serta perubahan histopatologi ginjal. Penelitian ini bertujuan menganalisis pengaruh kandungan omega-3 buah merah Papua (Pandanus conoideus Lamk. ) terhadap kadar BUN, kreatinin, dan gambaran histopatologi ginjal pada tikus Wistar yang dipapar asap rokok. Penelitian menggunakan metode true experimental dengan rancangan post-test only control group design pada 25 ekor tikus putih jantan Wistar yang dibagi ke dalam lima kelompok, yaitu kontrol negatif (K-), kontrol positif (K+), dan tiga kelompok perlakuan (P1, P2, dan P3). Kelompok perlakuan memperoleh ekstrak buah merah Papua dengan dosis 0,3 ml/kgBB, 0,6 ml/kgBB, dan 0,12 ml/kgBB setelah pajanan asap rokok. Parameter yang diamati meliputi kadar BUN, kreatinin, dan histopatologi ginjal. Analisis data dilakukan menggunakan uji Shapiro–Wilk, Levene, Kruskal–Wallis, Mann–Whitney, dan Mood’s Median Test. Hasil penelitian menunjukkan bahwa pajanan asap rokok meningkatkan kadar BUN dan kreatinin serta menyebabkan kerusakan histopatologi ginjal. Sebaliknya, pemberian ekstrak buah merah Papua menurunkan kadar BUN dan kreatinin serta memperbaiki struktur jaringan ginjal pada seluruh kelompok perlakuan. Efek terbaik terhadap penurunan BUN ditemukan pada kelompok P2, sedangkan penurunan kadar kreatinin dan perbaikan histopatologi ginjal terbaik terlihat pada kelompok P3. Dengan demikian, ekstrak buah merah Papua berpotensi sebagai agen nefroprotektif terhadap kerusakan ginjal akibat pajanan asap rokok.
