Abstract
Cyclooxygenase-2 (COX-2) is an enzyme that plays a role in prostaglandin synthesis in the inflammatory phase. COX-2 has an important function in the regulation of renal hemodynamics and COX-2 can be affected by anti inflammatories. One of the herbs that can be used for anti-inflammatory is berenuk fruit. The purpose of this study was to determine the distribution of COX-2 in the kidney organ by immunohistochemistry after giving berenuk fruit fermentation. This study used 18 Sprague Dawley rats divided into 3 treatments and 6 replicates. The treatment groups were P1: as control, P2: fermented berenuk fruit dose of 2.96 mg/kg BW, P3: fermented berenuk fruit dose of 5.92 mg/kg BW. Therapy for 7 days was given using an oral sonde. Rats were cervically dislocated and kidney organs were removed on day 8. The kidneys were processed histopathologically with immunohistochemical staining. Data were analyzed by One Way Anova test followed by Duncan's test. The results showed that there was no effect of treatment on COX-2 immunoreactivity in the kidneys, P = 0.06 (P > 0.05). COX-2 immunoreactivity is localized mainly in the renal tubular epithelium and glomerulus is not immunoreactive to COX-2. Cell nuclei were seen evenly distributed throughout the tissue, but were not a major indicator of COX-2 expression. The conclusion of this study is that the fermentation of berenuk fruit has an effect on reducing COX-2 expression in Sprague Dawley rat kidney tissue qualitatively, but not statistically different.
