Abstract

ABSTRAK Penelitian ini dilakukan untuk mengetahui toksisitas akut ekstrak seledri (Apium graveolens) terhadap histopatologi lambung tikus (Sprague dawley). Dalam penelitian ini menggunakan 24 ekor Tikus putih jantan galur Sprague dawley sebagai hewan percobaan dengan berat badan 200-250 gram. Tikus dibagi kedalam 4 kelompok perlakuan yaitu kelompok P0 sebagai kontrol, kelompok P1 diberi ekstrak seledri dengan dosis 50 mg/kgBB, kelompok P2 diberi ekstrak seledri dengan dosis 500 mg/kgBB, kelompok P3 diberi ekstrak dengan dosis 5000 mg/kgBB dilakukan selama 14 hari dengan cara peroral menggunakan oral sonde sehari 1 kali. Tikus kemudian di eutanasia pada hari ke 14 dengan menggunakan metode dislokasi cervical. Nekropsi dilakukan untuk mengambil organ lambung yang kemudian dijadikan dalam bentuk preparat dengan pewarnaan HE. Lesi yang diamati yaitu inflamasi, nekrosis, hemoragi, degenerasi dengan menggunakan mikroskop. Hasil kemudian dianalisis menggunakan metode Kruskal Wallis untuk menentukan perbedaan pada kelompok kontrol dan perlakuan. Berdasarkan hasil penelitian yang telah dilakukan, memperlihatkan bahwa terdapat pengaruh perlakuan ekstrak seledri (Apium graveolens) dengan berbagai dosis terhadap histopatologi lambung tikus. Pada kelompok P2 dan P3 pemberian ekstrak seledri (Apium graveolens) memiliki dampak inflamasi, nekrosis dan hemoragi yang nyata jika dibandingkan dengan kelompok P0 dan kelompok P1 (p≤0,05). Hal ini menunjukkan bahwa ekstrak seledri (Apium graveolens) pada dosis 500 mg/kgBB mulai menimbulkan efek toksisitas pada lambung. Kata Kunci : Toksisitas Akut, Seledri (Apium graveolens), Lambung ABSTRACT This study was conducted to determine the acute toxicity of celery extract (Apium graveolens) on gastric histopathology of white rats (Sprague dawley). In this study, 24 male white rats of the Sprague dawley strain were used as experimental animals with a body weight of 200-250 grams. The rats were divided into 4 treatment groups, namely group P0 as a control, group P1 given celery extract at a dose of 50 mg/kgBW, group P2 given celery extract at a dose of 500 mg/kgBW, group P3 given extract at a dose of 5000 mg/kgBW for 14 days orally using an oral probe once a day. The rats were then euthanized on day 14 using the cervical dislocation method. Necropsy was performed to take the gastric organs which were then made into preparations with HE staining. The observed lesions were inflammation, necrosis, hemorrhage, degeneration using a microscope. The results were then analyzed using the Kruskal Wallis method to determine differences in the control and treatment groups. Based on the results of the research that has been done, it shows that there is an effect of celery extract (Apium graveolens) treatment with various doses on the gastric histopathology of rats. In groups P2 and P3 administration of celery extract (Apium graveolens) had a significant impact on inflammation, necrosis and hemorrhage when compared to groups P0 and group P1 (p≤0.05). This indicates that celery extract (Apium graveolens) at a dose of 500 mg/kgBW started to cause a toxic effect on the gastric. Keywords: Acute Toxicity, Celery (Apium graveolens), Gastric
